Effects of Connective Tissue Growth Factor on the Cell Viability, Proliferation, Osteogenic Capacity and mRNA Expression of Stem Cell Spheroids

Background: Connective tissue growth factor (CTGF) is a cellular communication network family protein involved in many functions. The purpose of this study was to determine the effects CTGF on proliferation, osteogenic capacity, and mRNA expression spheroids composed gingiva-derived mesenchymal stem cells (GMSCs). Methods: applied at final concentrations 0, 25, 50, 100, 200 ng/mL. Qualitative cell viability determined using Live/Dead kit assay. Metabolic with colorimetric assay kit. Osteogenic activity analyzed alkaline phosphatase Alizarin Red S staining. Quantitative polymerase chain reaction (qPCR) used assess levels RUNX2, BSP, OCN, COL1A1. Results: In general, there no significant difference between groups Days 1, 4, 7. Addition produced an increase qPCR results demonstrated that COL1A1 were significantly increased addition CTGF. Conclusions: Based these findings, we conclude can be for differentiation spheroids.
